چکیده

Background Primary Sjögren’s syndrome (pSS) is an autoimmune disease characterized by B cell hyperactivity. CXCR5+ follicular helper T cells (Tfh), CXCR5-PD-1hi peripheral (Tph) and CCR9+ Tfh-like have been implicated in driving hyperactivity pSS (1-3), however, their potential overlap has not evaluated. Objectives To study the between two CXCR5- subsets to PD-1/ICOS expression, hall mark markers, as compared “true” CXCR5/PD-1/ICOS-expressing Tfh cells. Methods We defined Tph memory (CD45RO+), CXCR5-CCR9+, CXCR5+CCR9+ CD4 cells, previously described. co-expressing were specified on phenotype. memory, CXCR5+PD-1+ICOS+ populations from blood mononuclear of patients (n=12) age- gender-matched healthy controls (HC, n=11) flow cytometry. expression these was each other taking into account differentiation status. Results In number significantly increased HC (median 0.53% versus 0.23% p=0.049). Also elevated HC, i.e. approximately 2.7% 1.7% (medians, p=0.019). CCR9 equally distributed Only a modest 2.1% (1.2-3.2%) expressed with interquartile range), indicating limited overlap. (Tfh) highest frequency PD-1hi either or without co-expression (PD-1hi differed CXCR5-CCR9- CXCR5-CCR9+ both p<0.0001.) No difference seen CXCR5+CCR9- (p=0.57). Within populations, expressing CXCR5 and/or showed trend higher ICOS HCs, but this only significant (28.6% 55%, medians for respectively, p=0.02) CCR9. However, found CXCR5/CCR9 which are enriched circulation patients. PD-1, ICOS, than CCR9- (in all three comparisons p<0.05). Conclusion distinct that can drive pSS. The known upregulated CCL25 CXCL13, ligands CXCR5, at inflammatory sites suggests concerted action facilitate migration frequencies positive Hence, effector may contribute ongoing immune responses References [1]Chen W, Yang F, Xu G, Ma J, Lin J. T Helper Cells Profile and CD4+CD25+Foxp3+Regulatory T Cells in Polycystic Ovary Syndrome

Background: Polycystic ovary syndrome (PCOS) is considered as the most common cause of female infertility that affects 4-10% of women in the reproductive age. Previous studies have shown the role of a balanced immune response in a successful pregnancy and fertility. Objective: To investigate the T helper cells type 1 (Th1) /Th2/Th17/Treg paradigms in peripheral blood of infertile PCOS compared ...

CD4+ T Cells are Exhausted and Show Functional Defects in Chronic Lymphocytic Leukemia

Background: Chronic lymphocytic leukemia (CLL) is the most common adult leukemia in the western world. This health problem is caused due to the accumulation of mature B-lymphocytes in the peripheral blood and bone marrow. In the course of cancer, CD4+ T cells become “exhausted” and characterized with poor effector functions and the expression of multiple inhibitory receptors. Objective: To inve...

CD4+ T Cells Expressing Latency-Associated Peptide and Foxp3 Are an Activated Subgroup of Regulatory T Cells Enriched in Patients with Colorectal Cancer

Latency-associated peptide (LAP) - expressing regulatory T cells (Tregs) are important for immunological self-tolerance and immune homeostasis. In order to investigate the role of LAP in human CD4⁺Foxp3⁺ Tregs, we designed a cross-sectional study that involved 42 colorectal cancer (CRC) patients. The phenotypes, cytokine-release patterns, and suppressive ability of Tregs isolated from periphera...

Eomesodermin-expressing T-helper cells are essential for chronic neuroinflammation

Development of acute experimental autoimmune encephalomyelitis (EAE) depends on Th17 cells expressing the nuclear factor NR4A2. However, in mice lacking NR4A2 in T cells, a late-onset disease is still inducible, despite a great reduction in acute inflammation. We here reveal that development of this late onset disease depends on cytotoxic T-cell-like CD4(+) T cells expressing the T-box transcri...

CD4+ Primary T Cells Expressing HCV-Core Protein Upregulate Foxp3 and IL-10, Suppressing CD4 and CD8 T Cells

Adaptive T cell responses are critical for controlling HCV infection. While there is clinical evidence of a relevant role for regulatory T cells in chronic HCV-infected patients, based on their increased number and function; mechanisms underlying such a phenomena are still poorly understood. Accumulating evidence suggests that proteins from Hepatitis C virus can suppress host immune responses. ...
